Apart from accidents and illness, scientists have discovered molecules at the end of chromosomes called telomeres that get shorter every time a cell divides. They think that every cell is programmed to die at somepoint, and when sufficient of them die, we die. They are researching this area to try to answer exactly your question and to see if it can be stopped.
